The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing Fth1. The ELISA analytical biochemical technique of the MBS288729 kit is based on Fth1 antibody-Fth1 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect Fth1 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, Fth1. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Intended Uses: This immunoassay kit allows for the in vitro quantitative determination ofmouseFerritin H subunitconcentrations in serum, Plasma,tissue homogenates and Cell culture supernates and Other biological fluids.

Principle of the Assay: The microtiter plate provided in this kit has been pre-coated with an antibody specific to Ferritin H subunit. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated polyclonal antibody preparation specific forFerritin H subunit and Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then a TMB substrate solution is added to each well. Only those wells that contain Ferritin H subunit, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The concentration of Ferritin H subunit in the samples is then determined by comparing the O.D. of the samples to the standard curve.

Ferritin-H: Stores iron in a soluble, non-toxic, readily available form. Important for iron homeostasis. Has ferroxidase activity. Iron is taken up in the ferrous form and deposited as ferric hydroxides after oxidation. Also plays a role in delivery of iron to cells. Mediates iron uptake in capsule cells of the developing kidney. Oligomer of 24 subunits. There are two types of subunits: L (light) chain and H (heavy) chain. The major chain can be light or heavy, depending on the species and tissue type. The functional molecule forms a roughly spherical shell with a diameter of 12 nm and contains a central cavity into which the insoluble mineral iron core is deposited.
